The System

Mechanical Lysis System

The microCompass™ System provides sound and accurate data based on molecular biology that allows an operator to achieve results in a 4 hour time frame. It offers many advantages such as automation, quick time to results, little or no pre-enrichment of original sample and reproducible results. The expected sensitivity of the system is 100 cfu per sample or 1 cfu/ml with a 100 ml sample. The microCompass system includes the following instruments: Mechanical Lysis System, RNA Extraction Robot and R.A.P.I.D.® LT Thermal Cycler.




RNA Extraction Kit and RNA Extraction Robot

The microCompass RNA Extraction Kit is suitable for the extraction and purification of total RNA from bacteria, yeast and mold. Product samples are filtered through the microCompass Spin Filtration Unit, then RNA Booster is added to equilibrate the ribosomal RNA pool. Microorganisms are lysed using a combination of mechanical and chemical lysis. Total RNA isolation is automated on a robotic platform using coated paramagnetic beads. Finally, the purified total RNA is eluted into a 96-well microplate, ready for real time RT-PCR. The RNA Extraction Kit includes Sample Lysis Buffer, Binding Buffer, Lyophilized DNase, DNase Buffer, Wash Buffers, Elution Buffer and microCompass™ Lysis tubes.




Detection Kits and the R.A.P.I.D. LT-Thermal Cycler

The microCompass™ Detection Kits contain all reagents for universal detection of bacterial or yeast and mold RNA. Each kit is used with Tth DNA polymerase, a thermo stable enzyme with RNA dependent reverse transcriptase activity and DNA dependent polymerase activity, allowing the combination of reverse transcriptase (RT) and PCR in a single tube reaction. Amplicons are detected using a MGB™ Eclipse specific probe and detected by monitoring the resulting fluorescent signal. The sensitivity is 50 fg of RNA. Total assay sensitivity (including sample pretreatment and RNA extraction) depends on the initial sampling volume.


The method is based upon detection of universal sequences of RNA (bacteria and spores) using a one-step Real Time Reverse Transcriptase PCR assay and MGB™ Eclipse probe technology. Universal sequences detected are based upon ribosomal 16S rRNA (bacteria) and 18S rRNA (yeast and molds). Ribosomes play a major role in protein synthesis and the detection of these RNA reflects protein synthesis and viability of microorganisms. The Detection Kits include Buffers, Primers and dehydrated RNA Positive Control and the Tth enzyme is supplied separately from the kit.
