Why are the plates left undisturbed for 3 weeks after transfection (isn’t there a risk of the medium evaporating, will the cells be in good condition, etc.)? |
 | | It is recommended to leave the plates completely undisturbed during the first 3 weeks following transfection (although one plate should be selected to check, at regular intervals, for contamination and the progress of the transfection). It has been found that this increases the number of colonies that are obtained. If the incubator is suitably humidified then evaporation should not be a problem. If there is concern about evaporation then it may be advisable to monitor the level of medium and, as an emergency measure, feed the wells with glutamine-free medium. In our experience, the cells do remain healthy for at least 3 weeks when left undisturbed like this. |
